How to add glide to one step on the Mother-32
On the Mother-32, glide is switched on or off for each step of a pattern, so you choose which notes the line slides into. The GLIDE knob does two jobs: while you are editing a step, turning it right switches that step's glide on; once you leave the edit, it sets how fast every gliding step slides.

Put the sequencer in Step mode (hold SHIFT and press the arrow-shaped STEP button) and start a pattern playing whose steps 2 and 3 are different notes: to change a step's note, hold SHIFT and press its step button, play a key, then press both again. Have the SUSTAIN switch on, so gate lengths sound as you set them, and GLIDE all the way left, as in the default settings.
Three steps
Press SHIFT + 2. Turn GATE LENGTH fully right. Step 2 now holds into step 3.

Good to know
To take glide off a step, edit the step again and turn GLIDE all the way left. The same step edit sets a step's note, gate length, accent, rest and ratchets; a gate length all the way right ties the step to the next one.
